The gold standard in anti-aging and acne management, retinoids operate via direct interaction with cellular DNA.
The Conversion Pathway
For a retinoid to exert biological activity, it must be converted into retinoic acid to bind to Retinoic Acid Receptors (RARs) and Retinoid X Receptors (RXRs) in the nucleus.

Mechanism of Action
Upon binding to nuclear receptors, retinoic acid initiates transcription of target genes, resulting in:
- Normalized Desquamation: Prevents microcomedone formation.
- Collagen Synthesis: Upregulates Types I and III collagen while inhibiting matrix metalloproteinases (MMPs) that degrade collagen.
- Epidermal Thickening: Reverses mucosal atrophy associated with photoaging.
- Melanin Dispersion: Accelerates the turnover of pigmented keratinocytes.
The "Retinization" Period
Initial use frequently causes irritant contact dermatitis, characterized by erythema, scaling, and burning. This is usually transient (2-4 weeks). Mitigation strategies include:
- The "sandwich method" (moisturizer - retinoid - moisturizer).
- Short contact therapy (applying for 30 minutes, then washing off).
- Gradual dose escalation (starting 1-2x per week).
